Abstract
Background
Cellular senescence is a condition in which the cells permanently stop dividing due to various conditions like DNA damage, shortening of telomeres, oxidative stress, and mitochondrial problems. Initially this step is considered to be protective because it prevents damaged cells from becoming cancerous, but later, as the age of the person increases, this senescence starts accumulating in the body and causes various problems in the body. These cells also release inflammatory substances, which are known as senescence-associated secretory phenotypes, which are responsible for chronic inflammation and may contribute to various age-related factors.
Purpose
The review aims to summarize the molecular mechanism, Chemistry of cellular senescence, its role in chronic diseases, pharmacological approaches of senolytic drugs.
Materials and Methods
Various papers were searched using Google Scholar and PubMed for senolytic, cellular senescence, dasatinib, quercetin, fisetin, rapamycin, navitoclax, the mechanism of senescence, chemistry, and clinical trials involved in this topic.
Results
this review discusses the molecular mechanism of cellular senescence, chemistry its role in chronic diseases, and also current pharmacologic approaches.
Conclusion
Targeting cellular senescent shows a promising therapeutic strategy, however further studies are required to established long term safety, efficacy and clinical translation
