Abstract
Background
Shatavari or Asparagus racemosus Wild. Family Liliaceae is an important therapeutic herb in the health care system. It contains a sapogenin content which is a precursor of many pharmacologically active steroids. This plant is used to induce menstruation and as a cosmetic tonic.
Objectives
The objective of the present study was to carry out a pharmacognostic and phytochemical evaluation of Asparagus racemosus Willd., with particular emphasis on the isolation, identification, and characterization of saponin glycosides using advanced analytical techniques such as HPTLC and LC-MS.
Materials and Methods
The roots of Asparagus racemosus were authenticated and extracted using a hydro-alcoholic solvent. Pharmacognostic, physicochemical, and preliminary phytochemical evaluations were performed using standard methods. HPTLC and LC-MS analysis were carried out to identify and characterize saponin glycosides, with particular emphasis on Shatavarin IV.
Results and Discussion
The evaluation revealed both external and internal diagnostic features of the root. Phytochemical analysis confirmed the presence of carbohydrates, glycosides, saponins, tannins, and steroids, with total saponin content of 20% w/w. Physicochemical parameters showed total ash (2.50% w/w), water-soluble ash (1.20% w/w), acid-insoluble ash (0.35% w/w), hydro-alcoholic extractive value (31.50% w/w), and loss on drying (4.73% w/w). HPTLC analysis revealed fifteen peaks (Rf 0.01-0.96) and confirmed the presence of Shatavarin IV. LC-MS analysis further identified saponin glycosides, with Shatavarin IV as the major compound at a retention time of 2.7 min.
Conclusion
The study establishes standardized pharmacognostic and physicochemical parameters for Asparagus racemosus and confirms the presence of bioactive phytoconstituents, particularly saponin glycosides. The detection and characterization of Shatavarin IV through HPTLC and LC-MS validate the authenticity and quality of the plant material. These findings support the therapeutic potential of A. racemosus and provide a scientific basis for its standardization, and pharmacological investigations.
