Abstract
Background
Shatapushpadi Lepa is a traditional external application specified in Ayurveda. The composition that is advised for Vata-dominant inflammatory joint diseases. To determine quality, and therapeutic effectiveness, such a conventional formulation must be scientifically validated.
Purpose
The present study is aimed to evaluate the quality of shatapushpadi lepa in inflammatory joint diseases like Rheumatoid arthritis through physiochemical, phytochemical profiling and Chromatographic analysis.
Objectives
To evaluate the quality of Shatapushpadi Lepa through comprehensive assessment of physiochemical, phytochemical profiling and Chromatographic analysis.
Material and Methods
The raw drugs were obtained from a GMP-certified Ayurveda pharmacy and authentication and analysis was done in an AYUSH-approved laboratory. Shatapushpadi Lepa was prepared in accordance with classical reference Organoleptic and physicochemical characteristics such as ash values, extractive values, moisture content, and pH were evaluated in accordance with API standards. Aqueous and alcoholic extracts were used to carry out phytochemical screening. Toluene was used for the Thin Layer Chromatography (TLC) study. The mobile phase consists of ethyl acetate and formic acid (3:6:1). To detect bioactive compounds, GC-MS analysis was performed on a Shimadzu GCMS-QP 2010SE.
Results
All raw drugs and the finished formulation complied with in-house quality requirements. The formulation had low ash values, a high moisture content (66.20%), and a slightly acidic pH (5.226), indicating its stability and appropriateness for topical application. Phytochemical research indicated the presence of flavonoids, tannins, glycosides, and reducing sugars, all of which have anti-inflammatory and antioxidant properties. TLC examination found many well-resolved spots with a high Rf value of 0.63, indicating a variety of phytoconstituents. GC-MS analysis revealed bioactive substances such triterpenes, apiol, carvone, longifolene, α-terpineol, and pentacosane that have anti-inflammatory, analgesic, antioxidant, and penetration-enhancing activities.
Conclusion
This study illustrates the quality control for Ayurvedic products, ensuring their reliability and therapeutic value. The compounds which are present in formulation shows anti-inflammatory, analgesic, and antioxidant effects, as indicated by pharmacological tests such as phytochemical, physiochemical, TLC, and GCMS analysis. Future study should focus on bioavailability and clinical effectiveness to further substantiate the therapeutic potential of this formulation.
